External Boot in my image

Hi, i have a sd card with retropie (original image from here with my favorite games) with 16gb, but i backup with "win32diskimage" and flash in external hard drive (to put more games and still my original config) but not boot in first time, after problem i change the cmdline.txt in external hard drive in boot (fat partition) to boot in sda2 (root=/dev/sda2) and inicial boot work but still try acess /dev/mmcblk0p2 (in verbose screen), if i put a empyt sd card the boot work and a lot of files save in this card if not insert card the rpi restart.
i try to find any information to change the boot in another file but i not find and i not know linux commands/files to change, any tip or help?tecnical information

raspberry pi 3
my rpi3 is set to boot external (tested with other images without sd card)
sd card 16gb
external hard drive 250gb (not work with my backup image but boot if flash a clean image :( )sorry my english, is rusty...

@MPlay Is it just a storage space issue? I run the OS on a 32gb card and keep my roms and saves on an external hard drive so it is easier and faster to copy/backup data.
If this is the case, try following the Automatic Mount instructions in the docs:

Thank you for tips, but i found solution, i edited cmdline.txt in fat partition (changed to sda2) and in linux via live cd i edited fstab in etc folder and change information to sda1 and sda2 and working, now the system boot direct in external hdd without sd card and load speed is great.
case resolve.

you are welcome, my next step is set retropie to stream my game play (if possible in net play mode) in my youtube channel.
