      I am confused as to which settings I need to use to config audio out of my speakers that I have tied to a mono amp , which is wired to the GPIO header. I have confirmed that the speaker works as far as testing if it plays audio files.

      My question is, what settings should I use, under "Sound Settings" in emulationstation?

        @Propellent The sound settings in Emulationstation apply only to the audio produced by Emulationstation, not the emulators. Have you tried using the 'default' Audio card ?
