ROMs do not launch

RetroPie set up correctly, ROMs running great, no problem. Then (seemingly without any settings changes) ROMs now will not launch.
EmulationStation runs, and can browse ROMs, but when it comes to launching ROMs it hangs for 2 seconds and then takes you back to the menu. The normal RunCommand box doesn't appear. This is the case for all games on all platforms/emulators.
I have completely uninstalled and reinstalled Retropie, updated everything, tried local ROMs and those on an HDD and yet it still doesn't work! Any ideas?
Pi Model or other hardware: RPI 4 Model B
Power Supply used: Mains through approved cable
RetroPie Version Used: 4.5.14
Built From: Installed through the GitHub tutorial
USB Devices connected: HDD
Controller used: 8BitDoSF30 Pro

UPDATE:
Trying to launch something from the 'Configuration' menu within RetroPie also does not launch anything, it exits as if it is going to and then returns to the menu.

Did you modify any settings for Runcommand (like disabling the menu) ? Do you have enough free disk space available ?

No Runcommand settings were changed before the problem started (I have since uninstalled and reinstalled RetroPie twice and checked the Runcommand settings and everything was set as it was as default).
There should be plenty of disk space available - around 4gb.

Try to turn on verbose logging from the Runcommand launch menu, then post the complete
/dev/shm/runcommand.log
file on pastebin.com. -
@mitu Thanks - because the Runcommand launch menu won't open (when I select a ROM it shows the desktop as normal but then just goes back to the menu) I can't switch verbose logging on.
I've had a look around for a way to switch on verbose logging elsewhere but haven't had any luck.

@grzzlybr EmulationStation should not be running from a Desktop session - the manual installation makes it clear you need to be running a Rapbian Lite.

I have just tried running RetroPie from boot and it seems to be working fine with local ROMs, but not with ROMs using symlinks (which were working fine before). So I guess that's one issue (sort of) solved and another found!

Sure, but that's not a configuration that's tested and supported.

Can you add more details - and possibly the log file asked earlier, not that Runcommand should be working again ?
