I am using a Genesis USB Adapter for the Retropie and it seems that it won't read the Up and Left buttons on the D-Pad.. VERY frustrating! Right and Down work but they work as Up & Down for some strange reason. Any help would be GREATLY appreciated!

      Thanks!!

You're not alone, it's simply the nature of the beast sadly:

Basically, I had to try different USB adapters with different controllers to find a couple that would work with RetroPie. Sorry I don't have a better solution.

@DRest608 try another controller. Even if it works in a real system. I also like the vision-daptor and ultimate pc interface. They work pretty good for genesis controllers. Adapters that have a metal male dsub as apposed to a plastic casing work better with genesis controllers as well. They fit tighter giving a better connection. @Dochartaigh i should have mentioned this last part to you.

Like this? ;) That's the RetroUSB one I've had the best luck with (albeit still NOT 100% reliable at all).

@Dochartaigh yes. A lot of the genesis cables have loose pins and these style of connectors generally have a tighter fit.
